No. Only PS/2 protocol. Work all USB mouses with ps/2 protocol support. Or can be used PS/2 connector.

At 100% support PS/2 protocol all USB mouses sold with USB>PS/2 adapter in package. Also part of original USB mouses contain internal controller compatible also with PS/2 protocol. K-MOUSE2016 pcb is designed for both connectors USB and PS/2, each user can select one.

the connector in the oposited side as the label "K-MOUSE2016" --- ¿? I can't imagine the propurse of it... maybe video output, audio in or out, remote NMI button
and some resistors in the expansion port...

as allways than I see your designs, I like it a lot
I saw the pin of the USB/PS2 mode in the corner (down-left)
I think only in one thing, the strange place for the led.

Values of some resistors can be different for different CPLD firmware. And big part of resistors work only as pull-up, then here can be any usable value.

Cinch is video out for ZX48 models. Throught connector contain some resistors, but can be removed for use with next firmwares (in future)....

In ESXDOS system is possible set joystick mode on K-MOUSE 2016 thanks to this commands (unofficial):

(all commands must be copied to directory "BIN" on your disc/card with system ESXDOS)

Commands for master K-MOUSE:
.km1_0 - disable joystick
.km1_1 - set fuller joystick
.km1_2 - set kempston joystick (a-mouse)
.km1_3 - set joypad+kempston

Commands for slave K-MOUSE:
.km2_0 - disable joystick
.km2_1 - set fuller joystick
.km2_2 - set kempston joystick (a-mouse)
.km2_3 - set joypad+kempston

For testing mouse can be used old tester for K-MOUSE 2011 because K-MOUSE 2016 is compatible:
